FORMULASI DAN OPTIMASI NANOSTRUCTURED LIPID CARRIERS (NLC) KETOKONAZOL MENGGUNAKAN FULL FACTORIAL DESIGN FORMULATION AND OPTIMIZATION OF NANOSTRUCTURED LIPID CARRIERS (NLC) KETOCONAZOLE USING FULL FACTORIAL DESIGN

Abstract
Ketokonazol merupakan antijamur golongan imidazol berspektrum luas. Ketokonazol masuk dalam Biopharmaceutics Classification System (BCS) kelas 2 yang memiliki kelarutan rendah dan permeabilitas tinggi. Pengobatan topikal obat Ketokonazol memiliki kelemahan yaitu rendahnya bioavalabilitas dan permeasi obat sehingga diperlukan formulasi dan optimasi sistem penghantaran Ketokonazol. Sistem Nanostructured Lipid Carrier (NLC) memiliki keuntungan untuk meningkatkan stabilitas fisikokimia bahan aktif, dapat menghidrasi kulit secara in vivo karena memiliki oklusifitas yang baik, dan meningkatkan skin targetting. Pada penelitian ini dilakukan studi pengaruh rasio lipid padat gliseril monostearat dan lipid cair coconut oil terhadap karakteristik fisikokimia dari NLC Ketokonazol menggunakan model full factorial design, untuk mendapatkan formula optimum dengan variabel bebas komposisi lipid padat dan lipid cair dengan variabel terikat ukuran partikel, Zeta potensial dan daya sebar. Pada penelitian ini didapatkan hasil bahwa ukuran partikel NLC Ketokonazol memiliki rentang 458,1± 0,6 hingga 900,9 ± 0.2 nm, dengan p value 0,04 (p < 0,05). Daya sebar dari NLC Ketokonazol memiliki rentang 4,8 ± 0,1 sampai 7,2 ± 0,1 cm, p values 0,707 (p > 0,05). Zeta Potensial dari NLC Ketokonazol memiliki rentang -32,1 ± 0,3 sampai -65,4 ± 0,8 mV, dengan p value 0,143 (p > 0,05). Rentang pH yang dihasilkan adalah 5,9 ± 0,2 hingga 6,4 ± 0,2, viskositas yang dihasilkan adalah 690 ± 0,7 hingga 833 ± 0,8 cps. Rasio konsentrasi lipid padat dan lipid cair berpengaruh signifikan pada ukuran partikel, tetapi tidak pada daya sebar dan zeta potensial
